Acid Dentin Lysates Increase Amelotin Expression in Oral Epithelial Cells and Gingival Fibroblasts

چکیده
Amelotin (AMTN) is a secretory calcium-binding phosphoprotein controlling the adhesion of epithelial cells to tooth surface, forming protective seal against oral cavity. It can be proposed that signals released upon dentinolysis increase AMTN expression in periodontal cells, thereby helping preserve seal. Support for this assumption comes from our RNA sequencing approach showing gingival fibroblasts exposed acid dentin lysates (ADL) greatly increased expression. In present study, we confirm significantly and extend observation towards cell lineage by use HSC2 squamous TR146 buccal carcinoma lines. immunostaining revealed an intensive signal nucleus lysates. Acid mediate their effect via transforming growth factor (TGF)-β type 1 receptor kinase as antagonist SB431542 abolished fibroblasts. Similar what known fibroblasts, lysate Smad-3 phosphorylation cells. also respond AMTN-stimulating activity when adsorbed gelatin. When simulating regenerative approaches, enamel matrix derivative, TGF-β1, bone morphogenetic protein-2 caused robust SB431542-dependent HSC2. Taken together, show here uses TGF-β-depended signaling pathway support possibly maintaining

drugs on oral mucosa cells. A previous study showed that 5 μM of zoledronic acid (ZA), a nitrogen-containing bisphosphonate, is the highest concentration of this drug found in the oral cavity of patients under treatment. Therefore, in order to simulate an osteonecrosis clinical condition, the aim of this study was to evaluate the highest concentration of ZA applied on human epithelial cells (Ha...
متن کاملResponse of a co-culture model of epithelial cells and gingival fibroblasts to zoledronic acid.
Osteonecrosis of the jaw is an adverse effect of bisphosphonates. While the etiopathogenesis of this condition has been investigated, the interactions and effects of bisphosphonates on oral mucosa cells remain unclear. It is hypothesized that cell culture models, such as co-culture or three-dimensional cell culture models, can provide valuable insight. Therefore, the aim of this study was to ev...
متن کاملCalprotectin expression by gingival epithelial cells.
Calprotectin, a heterodimer of MRP8 and MRP14 with antimicrobial properties, is found in the cytosol of neutrophils, monocytes, and human gingival keratinocytes. During inflammation of the oral mucosa, the expression of immunoreactive calprotectin appears upregulated. Given the possible cell sources, we sought to learn if epithelial cells upregulate calprotectin in response to proinflammmatory ...
متن کاملDifferential cytotoxicity of long-chain bases for human oral gingival epithelial keratinocytes, oral fibroblasts, and dendritic cells
Long-chain bases are present in the oral cavity. Previously we determined that sphingosine, dihydrosphingosine, and phytosphingosine have potent antimicrobial activity against oral pathogens. Here, we determined the cytotoxicities of long-chain bases for oral cells, an important step in considering their potential as antimicrobial agents for oral infections. This information would clearly help ...
